Associate Scientist/ Scientist I – Antibody Discovery
The Antibody Discovery team at Tectonic is seeking a highly motivated and skilled Associate Scientist or Scientist I to aid in the discovery and engineering of innovative antibody therapeutics targeting GPCRs. Utilizing our novel GEODe technology, which enables the identification of functional antibodies to therapeutically relevant GPCR targets, this role will be central to our drug discovery pipeline. The ideal candidate will be an expert in utilizing antibody yeast surface display for naïve and affinity maturation campaigns, with a strong technical grasp of bio-layer interferometry (BLI) for detailed characterization of the binding kinetics of antibody hits. The candidate should be an independent and creative researcher, passionate about collaborating with a team at the forefront of molecular discovery, where their contributions will directly impact the development of next-generation GPCR targeting biologics. Key responsibilities to be successful in this role: Leading naïve and affinity maturation campaigns from large-scale yeast libraries using magnetic-activated cell sorting (MACS) and fluorescent-activated cell sorting (FACS). This will require the development and implementation of complex sorting and gating strategies to isolate rare, high-quality hits. Integration of next-generation sequencing (NGS), and subsequent data analysis, to track clone enrichment and diversity from yeast selection campaigns.Design and construction of next-generation yeast displayed naïve and affinity maturation antibody libraries.Utilizing BLI to perform high-throughput screening of hits and further detailed analysis of leads to characterize binding kinetics and map epitopes.Strong collaborations across groups for antigen design and downstream assays to ensure successful campaigns and thorough characterization of resulting clones. What you bring to the role: Ph.D. with 0+ years of post-graduate relevant laboratory experience, or BS/MS with 6+ years of relevant industry experience. Title commensurate with experience. Deep understanding of, and significant hands-on experience with yeast display platforms for the discovery of antibody therapeutics, including library preparation (cloning, DNA isolation and transformations) and multi-color flow cytometry/FACS. Experience with NGS for sequence analysis of selection campaigns is highly desirable.Proficiency in operating and troubleshooting BLI platforms with a solid understating of binding experiments for protein-protein interaction analyses including kinetics, competition and epitope binning. Compensation: Targeted Base: $100,000 - $155,000** Base Compensation for this role will depend on a number of factors including a candidate’s qualifications, skills, competencies, and experience, and may fall outside of the range shown. Base pay is only one component of the company’s total rewards package, all regular employees are also eligible for the corporate bonus program or the incentive compensation program (if applicable), as well as equity.
